Karen Shettler Paddock — Levaquin, Fluoroquinolone Toxicity & Spinal CSF Leaks

Chronology of Events
- October 1989 — Car accident: Karen suffered a serious car accident. Headaches, neck pain and other symptoms began during this period.
- 1993: Marriage to Bob Paddock. Symptoms continued and gradually worsened.
- 2004 — Levaquin (levofloxacin) prescribed: A fluoroquinolone antibiotic was given. According to Bob, symptoms associated with fluoroquinolone exposure emerged approximately six months later.
- 2004 and beyond — A long, difficult road: Years passed before the connection to the antibiotic was understood. Documented issues included fluoroquinolone toxicity, systemic tendinitis, intracranial hypotension, chronic pain and life-changing disability.
- 2012 — Spinal CSF leak diagnosis: After evaluation by 35 doctors, Karen received a diagnosis of a possible spinal CSF leak. Multiple procedures followed (epidural blood patches, lumbar punctures, surgery). Rebound high pressure and persistent low-pressure symptoms continued.
- 7 August 2013: Overwhelmed by pain and hopelessness after more than twenty years of symptoms, Karen died by suicide.
The Connection That Deserves Serious Attention
Karen’s published journal explicitly describes fluoroquinolone antibiotic poisoning from Levaquin (levofloxacin) and systemic tendinitis as part of her medical history.
Bob has spoken about the fluoroquinolone–CSF leak connection within the CSF leak community, including at the first CSF Leak conference. He has heard directly from 63 people who report that their own CSF leak began after taking a fluoroquinolone antibiotic.
Fluoroquinolones are known to cause serious connective-tissue injuries, including tendinitis and tendon rupture. The spinal dura is connective tissue. Connective-tissue weakness is a recognised risk factor for spontaneous CSF leaks.
Sixty-three patient reports should not be ignored. The fluoroquinolone–CSF leak connection deserves serious attention, careful documentation and dedicated scientific research.
Bob Paddock: Carrying Karen’s Legacy Forward
After Karen’s death, Bob was left with more than 9,000 journal entries, diaries and emails. He compiled them into a website and a book so that others could benefit from her experiences. Today he continues advocating, speaking and educating others about CSF leaks and the possible connection with fluoroquinolones.
His mission is simple but powerful: make sure Karen’s voice keeps helping others.

Chronologie van de gebeurtenissen
- Oktober 1989 — Auto-ongeluk: Karen had een ernstig auto-ongeluk. Hoofdpijn, nekpijn en andere klachten begonnen in deze periode.
- 1993: Huwelijk met Bob Paddock. De klachten hielden aan en verergerden geleidelijk.
- 2004 — Levaquin (levofloxacine) voorgeschreven: Ze kreeg een fluorochinolon-antibioticum. Volgens Bob ontstonden de klachten die met fluorochinolonen in verband worden gebracht ongeveer zes maanden later.
- 2004 en daarna — Een lange, moeilijke weg: Er gingen jaren voorbij voordat het verband met het antibioticum werd begrepen. Gedocumenteerde problemen waren onder meer fluorochinolonentoxiciteit, systemische tendinitis, intracraniale hypotensie, chronische pijn en een invaliditeit die haar hele leven veranderde.
- 2012 — Diagnose spinaal liquorlek: Na onderzoek door 35 artsen kreeg Karen de diagnose van een mogelijk spinaal liquorlek. Er volgden meerdere ingrepen (epidurale bloedpatches, lumbaalpuncties, een operatie). Daarna bleven zowel rebound-overdruk als aanhoudende klachten van te lage druk bestaan.
- 7 augustus 2013: Overweldigd door pijn en hopeloosheid na meer dan twintig jaar klachten, overleed Karen door zelfdoding.
Het verband dat serieuze aandacht verdient
Karens gepubliceerde dagboek beschrijft uitdrukkelijk een vergiftiging door het fluorochinolon-antibioticum Levaquin (levofloxacine) en systemische tendinitis als onderdeel van haar medische geschiedenis.
Bob heeft binnen de gemeenschap van liquorlekpatiënten gesproken over het verband tussen fluorochinolonen en liquorlekken, onder meer tijdens de eerste CSF Leak-conferentie. Hij heeft rechtstreeks gehoord van 63 mensen die melden dat hun eigen liquorlek begon na het gebruik van een fluorochinolon-antibioticum.
Fluorochinolonen staan erom bekend dat ze ernstige schade aan bindweefsel kunnen veroorzaken, waaronder peesontsteking en peesruptuur. De spinale dura is bindweefsel. Bindweefselzwakte is een erkende risicofactor voor spontane liquorlekken.
Drieënzestig patiëntmeldingen mogen niet worden genegeerd. Het verband tussen fluorochinolonen en liquorlekken verdient serieuze aandacht, zorgvuldige documentatie en gericht wetenschappelijk onderzoek.
Bob Paddock: Karens nalatenschap voortzetten
Na Karens overlijden bleef Bob achter met meer dan 9.000 dagboekaantekeningen, notities en e-mails. Hij heeft deze gebundeld in een website en een boek, zodat anderen iets aan haar ervaringen kunnen hebben. Nog altijd zet hij zich in, geeft hij lezingen en informeert hij anderen over liquorlekken en het mogelijke verband met fluorochinolonen.
Zijn missie is eenvoudig maar krachtig: ervoor zorgen dat Karens stem anderen blijft helpen.
